The PCR annealing temperature is calculated using the following empirical formula:
\[ T_a^* = 0.3 \cdot T_{mp} + 0.7 \cdot T_{mt} - 14.9 \]
Where:
\( T_a^* \) = Optimal annealing temperature
\( T_{mp} \) = Melting temperature of the primer
\( T_{mt} \) = Melting temperature of the target DNA
